Level 4 · Tested in the physical world

LabGenius reports its machine-designed Nectin-4 x CD3 engager kills tumour cells at 0.7 pM and cuts tumour growth by over 90% in mice

Nectin-4 is already a drug target, and what limits it is the healthy tissue that also carries it; LabGenius's claim is that selectivity can be engineered by how many arms have to bind at once (three to one here, so only dense target expression fires the molecule) rather than by finding a better epitope, and that its own discovery engine is what found the design. This is a conference abstract: mice and cell lines, the company's own assays, no paper, and the dosing interval it proposes rests on preliminary pharmacokinetics.

Level 4 · Tested in the physical world

LabGenius' robotic loop built 980 T-cell engagers over five cycles and found ones that killed HER2-high cells at single-digit picomolar and left the low-HER2 model untouched

On-target damage to healthy tissue is what has kept T-cell engagers out of solid tumours, so a loop optimising the gap between two cell lines aims at the real obstacle. The selectivity figure is a floor, not a measurement: it comes from seeing nothing at the highest dose tested. Everything is cells in a dish, run in-house with two donors per lead, and the paper says these readouts may not match a body. The healthy model is one cell line, T47D, which the paper says carries about as much HER2 as cultured heart muscle cells.

Research entries are the timeline's graded entries that name this company. A level grades one claim, not the company, and a blank can mean the record has not caught up rather than that nothing was published.
